M phase specific arrest, vinca alkaloid
1) bind to tubulin
2) block tubulin polymerization
3) arrest and death
Vinblastine USES
testicular tumors (w/bleomycin & cisplatin) ABVD for Hodgkin's disease
Vinblastine Toxicity
myelosuppression, nausea, vomiting
Vincristine MOA

child ALL (w/ glucocorticoids) Hodgkin's & non-Hodgkin's lymphomas
Vincristine Toxicity
dose-limiting neurotoxicity (peripheral neuropathy)
relatively low toxicity in bone marrow
Paclitaxel MOA
taxane
1) bind to tubulin
2) BLOCK DEpolymerization/disassembly
3) no desegregation in mitosis
Paclitaxel USES
metastatic breast/ovarian, lung, head/neck
Paclitaxel Toxicity
peripheral neuropathy hypersensitivity rxns (b/c from yew trees?)
Docetaxel MOA
taxane
1) bind to tubulin
2) BLOCK DEpolymerization/disassembly
3) no desegregation in mitosis
Docetaxel USES and toxicity
hormone refractory prostate cancer
and same as paclitaxel
