What is the body? What does it include?
Main section of the microscope minus the base and illuminators. Includes the eyepiece and objective lenses but not focusing block.
What is the Coarse Focus?
The larger of the focusing knob, it adjusts the distance to the specimen in large steps.
Describe the diaphragm and what it does.
Five-hole disc housed under the stage to adjust the amount of light passing through the stage opening.
Describe the stage openings and their role.
Lets light pass through to properly illuminate the specimen, while increasing contrast and resolution.
The eyepiece is made up of what lenses and what magnification levels do they include?
Ocular lenses with magnifications of 5X, 10X ,15X, and 20X.
Which is the Fine Focus, what does it do, and when do you use it?
Smaller of the adjustment knobs that adjusts the distance from the specimen in small steps towards various parts of the a specimen.
What does the Nose Piece hold and what does it do?
Structure that holds the objective lenses. Can be rotated to switch the lense.
How can you identify the objective lens? Which is the scanning lens? What other lenses are there?
Closes lense to the object being viewed. With different lenses at different magnification: Scanning (4X), Low-power (10X), Mid-power (40X), and High-power lense (100X).
On/Off controls what?
Controls the power to the microscope.
What is the stage and what is its function?
Flat plate that holds the slides for observation.
What is the power of the typical eyepiece lens?
Typically 10X.
When would you use a 100X lens?
Within oil immersion.
What color is assigned to identify the resolution of each objective lens?
Red (4X), Yellow (10X), Blue (40X), White (100X).
How do you calculate magnification of the image? An eyepiece of 10x and objective lens of 40x gives what total magnification?
Multiply the power of the eyepiece by the power of the objective lense. 10X * 40X would be 400X total magnification.
What lens is typically used with oil? What does the oil do?
100X, The immersion oil increases their resolution.
Describe the process of using oil. The oil serves as what?
A drop of oil is place on on the specimen slide. The oil now serves as a bridge between the lens and the glass palte contianing the specimen helping to focus the light and increase the resolution.
When do you remove the dust cover?
Until the microscope is place and positioned on the table where it will be used.
Can you wipe the lens with your finger? Why not?
No, oil from your hands could smudge the lens of even scratch it.
What do you use to clean lens?
Lens tissues.
What do you do to microscope when prepared to store.
Nosepiece should be in its lowest power setting, powest position to the satge. and the power cord neatly wrapped up, the dust cover should be replaced.
How does a condenser lens help?
Helps increase illumination and resolution by focusing/condensing light onto the specimen.
What is the cover slip and what does it do?
Thin square plastic or glass sued to cover the specimen on a slide.
What is resolution?
A lens system’s ability to resolve find details of the observed object.
What is the stage adjustment knob.
Mechanical stage allows for adjustment of the slide position forward, backward, and left or right.
What does the X stand for in a lens with a 200X?
Represents the magnification factor.
