1
Q
What is the function of a stage micrometer
A
- to calibrate an eyepiece graticule so it can be used to make measurements
2
Q
How is an eyepiece graticule calibrated
A
- select the desired magnification and objective
- place the stage micrometer of stage
- line up the scales of the micrometer and the eyepiece graticule
- count the number of division on the eyepiece graticule equivalent to each division on the stage micromete
- calculate the length of one division of the eyepiece graticule
3
Q
Outline procedure preparing an onion slide
A
- use forceps to peel epidermal layers off the onion
- mount onto slide with a drop of water
- add drop of iodine solution on the slide
- lower the cover slip onto the miscrope slide using a mounted needle slowly prevent air bubbles from forming

Q
Outline procedure of focusing with a light micrscope
A
- rotate the revolving nosepiece to line up the lower power objective len
- look throguh the eyepiece and use coarse adjustment knob to lower tha stage to focus the image
- ue the fine adjustment knob in order to focus image further
