Microscopy Flashcards
What are the advantages of a light microscope?
Cheaper
What are disadvantages of a light microscope?
Low magnification
Low resolution
What are advantages of an electron microscope?
High magnification
High resolution
What are disadvantages of an electron microscope?
Expensive
What is resolution?
How blurry an image is
How do we calculate magnification?
Image size / real image size
What is the independent variable for microscopy rp?
Objective lens
What is the dependent variable?
Magnification
What is your control variable?
The cell
How do we prepare a slide?
Remove a thin layer of specimen and place on slide
Add few drops of iodine stain
Remove excess with paper towel
Lower cover slip onto sample at an angle to get rid of air bubbles
What are the stages for microscopy?
Place slide onto stage
Select lowest power objective lens
Position objective lens until it almost touches the slide
Turn coarse focusing dial
Look down eyepiece
Slowly turn coarse focusing dial to increase distance between lens and slide
Use fine focussing dial to make image focused
Use formula
What is the role of the coarse focusing dial?
To focus on the lower / medium power objective lens
What is the role of the fine focusing dial?
To focus on high power objective lens
When viewing the specimen for the first time which objective lens should a student use? Give a reason for your choice.
The lowest magnification to give the widest field of view
What is the formula for total magnification?
Magnification of eye piece lens x magnification of objective lens
