Microscopy Flashcards
What is magnification?
what is resolution?
magnification -how many times bigger
an image is compared to actual object size
Resolution - the ability to see two objects as separate entities (detail)
How do you calculate magnification ?
magnification:
size of image /size of real object
how do you do magnification calculation with a scale bar?
-Find magnification of scale bar by dividing the ‘ruler measurement’ by the size given of scale bar
-Then use that info with the formula triangle :
I : AxM
How do youconvert
from mm to um to nanometres?
x1000. x1000
mm > um > nm
/1000. /1000
Describe a light microscope
- light is sent from light source through a specimen, magnified by glass lenses
*living cells - Black and white image.
- Cheap
- easy to use
- 2D image
- convex lenses, long wavelength
Deschbe a TEM microscope
•electrons pass through specimen to create image
• 2D
• Black and White
• specimen stained + in vaccum
• High magnification and resolution
Describe an SEM microscope
• electrons bounce off surface of specimen
•3D
• black+ white but can false colour with computer
• High magnification and resolution
Describe a Laser scanning confocal microscope
• uses a high power beam of light
to create mage. (laser)
• 3D and 2D
• fluorescent
• expensive
•high resolution
What are the resolutions of
• light
•SEM
•TEM
microscopes ?
•light: 200
•SEM : 3 - 10
•TEM : 0.2 - 0.5
what are the magnifications of
- light
-SEM
- TEM mioroscopes ?
•light: x 1,500 - 2000
•SEM: x 100,000 - 500,000
•TEM: x 500,000 - 2,000,000
