why is mt resp dependant on ADP
coupling
electrochem grad from proton pumping causes temp store of enerfy to phosphorylate ADP via ATPsynthase
coupling means ATP only produced when needed - ie only when there is a supply of ADP
Why is the rate of respiration in the presence of succinate and ADP greater than with glutamate plus malate and ADP?
malate is NADH linked - so fed into NADH dehydrogenase in complex 1
succinate is FADH2 linked so enters through succinate dehydrogenase in complex 2
therefore more succinate must be oxidised than malate/glutamate
why is it necessary to add simultaneously glutamate and malate as NADH-linked substrates?
needed for malate-aspartate shuttle in vivo
oxaloacetate is low and malate is high because of malate dehydrogenase
if added malate alone - oxaloacetate would build up and inhibit malate dehydrogenase and so ATP producton
addition of glutamate allows oxaloacetate to be removed from matrix via shuttle
active shuttle also means a-ketoglutarate is made in amtiporter allowing constant flow of malate into mt
KCN
covalently bond wit Fe3+ in cytochrome oxidase
inhibit terminal step in etc
oligomycin
inhibits ATP synthase and so resp
oligomycin in uncoupled mt
krebs and ox phos run at max but lot linked to ATP production
oligomycin has no effect
energy lost as heat
large O2 consumption
What do the data allow you to conclude regarding the metabolic effect of DNOC?
aromatic acids eg DNOC and DNP in undissociated form and dissipate theh electrochem grad
Why were the respiratory rate and body temperature elevated?
etc runs uncontrollably
energy released as heat
absence of body fat
use triglycerides for resp, from fat stored
erythropoiesis and increased pul resp
excessive O2 consumption = hypoxia
symptoms are an attemot to alleviate this
