Cyclo-oxygenase 1 & 2 inhibitors
They inhibit the COX enzymes from producing prostaglandins, therefore reducing the inflammatory response in the body
What is the difference between COX 1 & COX 2 inhibitors?
COX 1 enzyme produces prostaglandins that activate platelets and protect the stomach lining whereas COX 2 enzymes do not. They both facilitate the inflammatory response.
What are the direct effects of COX 1 & 2 inhibition?
inhibition of COX‑1 results in impaired gastric cytoprotection and antiplatelet effects
inhibition of COX‑2 results in anti-inflammatory and analgesic action
reduction in glomerular filtration rate and renal blood flow occurs with both COX‑1 and COX‑2 inhibition.
What are the adverse effects of NSAIDs?
nausea, dyspepsia, GI ulceration or bleeding, raised liver enzymes (especially diclofenac), diarrhoea, headache, dizziness, salt and fluid retention, hypertension
How do corticosteroids work?
They inhibit gene expression that controls the release of proinflammatory mediators and elements of the uimmune response.
