Lincosamides Flashcards
MOA?
Act by reversibly binding to the 50S ribosomal subunit at the 23S portion of the subunit inhibiting bacterial biosynthesis
Spectrum of activity?
G(+)
What tier do these belong to?
Tier2/3
What are some indications to use these drugs?
Bone infections, periodontal disease, URTI, skin
What specific bacteria can they be useful against?
Penicillin-resistant Stap and strep, actinomyces, nocardia, myocoplasma, fusobacterium, bacteriodes
What are some mechanisms of resistance?
Active efflux mechanism in some bacteria, methylase enzyme which modifies ribosomal target, hydrolysis of macrolides by esterases, chromosomal mutation of ribosomal binding sites
What is the distribution?
Good bone penetration
Poor distribution in eye (unless meninges are inflamed)
What are some examples of these drugs?
Lincomycin and Clindamycin
What is lincomycin used for?
Used as a feed additive in poultry
What bacteria is clindamycin used against?
G(+) anaerobic bacteria, toxoplasma and neospora
