Leture 17 Flashcards
What is able to be identified with the naked eye?
frog egg
What is able to be identified with the light microscope?
plant cells
animal cells
bacterium
virus ribosomes, sometimes electron microscope would be best
What can be viewed only by a electron microscope?
globular protein
small molecules
atom
How many layers of magnification does a light microscope have?
three
What are four types of light microscopy?
Bright field
Phase Contrast
DIC
Dark field
What is Bright field microscopy?
light transmitted through specimen
What is phase contrast microscopy?
alterations in light transmitted as changes in brightness
- light that is bouncing off of structures causes phase contrasts, more details
What is DIC microscopy?
highlights edges where there are changes in refractive index
What is dark field microscopy?
specimen is lit from the side and only scattered light is seen
What type of tissue is sectioning made of?
Live tissue
How does sectioning tissue occur?
A section of tissue is taken and placed in wax or frozen. This block is then cut to make slides that are able to be stained and viewed on a microscope.
What are the types of tissue staining?
Hematoxylin and Eosin
What does Eosin stain?
basic/positive charge
What does Hematoxylin stain?
acidic/negative charge
What is used in fluorescence microscopy to attached to proteins?
flurophore
