Safety of the vectors

A

1)It has mutations in the genome for not be able to integrate the DNA (the integrase D64V mutation in Pol gene)
2) Multiple plasmids, no single plasmid contains all the viral genetic information, there’s no way for the full set of viral genes to be reassembled in the infected cell.
3) Incorporates a deletion in the U3 region of the 3′ long terminal repeat (LTR). This “self-inactivating” (SIN) design prevents the LTR from driving transcription after integration
