Lecture 8 C - Legionella pneumoPHILA (Dr. Daniels) Flashcards
Where was Legionella pneumoPHILA found?
PHILAdelphia
What disease is caused by Legionella pneumophila?
Legionnaire’s disease
Gram +/-?
Gram -
Describe L. pneumophila
gram negative
pleomorphic (many shapes)
obligate aerobe (needs oxygen)
needs CYSTEINE
ENCOUNTER
contaminated water
What is L. pneumophila is a ______ of protozoa
parasite
What kind of temp does it like?
thermotolerant (up to 46 degrees C)
Encountered through…
environmental aerosols
NOT person-to-person
Multiply/Spread
- phagocytosed in alveoli
- multiply w/in autophagosomes of phagocytic immune cells (alveolar macrophages)
- inflammation –> acute bronchopneumonia
Damage/Immune Response
- Cell mediated immune response
2. Virulence/Proinflammatory mechanisms
What occurs w/ cell mediated immune response to L. pneumophila?
secrete IFN-gamme (sequester Iron)
What are some proinflammatory/virulent mechanisms of L. pneumophila?
- survives in macrophages
- LPS has cytotoxic effects
- FLAGELLIN
What does Flagellin do?
It is recognized by TLR-5 receptors on innate immune system cells and therefore inhances inflammation.
How do we treat Legionaire’s disease?
Antimicrobial drugs with good INTRACELLUAR PENETRATION.
- macrolides
- fluoroquinolones
- tetracyclines
Can penicillin treat legionnaire’s disease?
No. it is polar, so it can’t cross the cell membrane well enough.
Which medications are used to treat legionnaire’s disease?
Macrolides
Fluoroquinolones
Tetracyclines
