Lecture 21 Flashcards
What are the synaptic sites of action for a drug that would affect NT function?
Pre-synaptic receptors
Post-synaptic receptors
Uptake
Metabolic enzymes
What is a common test model for anxiety in mice?
The elevated cross model
How much of the US population suffers from some form of anxiety?
28.5%
Give examples of anxiety disorders
Generalised anxiety disorder Panic disorder Phobias OCD PTSD
What is the main type of anxiolytic?
Benzodiazepines
How do benzodiazepines work in anxiety?
They bind to an alternative site on the GABAa receptor to potentiate the effect of GABA
Give an example of a benzodiazepine with sustained action
Diazepam (Valium)
Give an example of a benzodiazepine with shorter action
Lorazepam
Why aren’t benzodiazepines used to treat anxiety long term?
They are very rapidly acting however tolerance and dependence builds quickly over 2 weeks
What are the effects of benzodiazepines?
Reduces anxiety and aggression
Sedation and induction of sleep
Reduction of muscle and liver co-ordination
Can cause amnesia
What is a commonly used long term treatment for anxiety?
Anti-depressants
What are the main class of hypnotics?

Give an example of a hypnotic benzodiazepine
Nitrazepam
Give an example of a non-benzodiazepine hypnotic
Zalepan (sonata)
Give an example of a non-bzd anxiolytic and how does it work?
Buspirane (Busper) which is a serotonin receptor agonist
What is commonly used to treat the side effects of the stress in anxiety?
Beta blockers e.g. Propanolol
What are rhe 2 main classes of therapeutic drugs in the CNS?
Neurological (brain)
Psychotropic (mind)
