Lecture 07-08 Flashcards
what are the function of benzos classified as
positive allosteric modulation of GABAa receptors (ligand-gated chloride ion channels)
hyperpolirizes the memrane, makes it harder to fire
pharmacodynamics
study of the effect of drugs on the body (ex. sleepiness, kills bacteria)
Benzo postive effects
sedation, sleep induction, anticonvulsant, muscle relaxtion, general reduction of signal transmission
efficacy of benzos
reduced anxiety symptoms in first two weeks , but no long term benefit
adverse benzo effects
drowziness, lack of coordination, concentration, decreased libido, ED, depression, disihnhintion, hypotension
How does the tolerances of benzos work
overtime the body will notice the high levels of GABAa receptors working and will modify gene expression to stop producing as many receptors, which thus causes a result ina. reduction of overall Chlorine channel openings
benzo withdrawl
due to tolerance issues overtime, benzos are needed to maintain and keep the same level, less inhintion of anxiety-evoking thoughts and events
Long term side effects of benzos
greater than two weeks,
Cogntive impairment, inc anxi/depression, anhedonia, altered perception of self/relationships
When are benzos prescibed?
gen anxiety/social anxiety disorder
usually reserved for pateints that have not responded to three treatemnts
acute emotional trauma, PTSD
How do barbiturates work
Posituve allosteric modualtion of GABAa, with additional antagonism of NMDA NA+ channels
at high doses can stimulate GABAa recptors directly
History of barbiturates
seen as huge breakthrough for anxiety and seizures, extremely addictive and fatal.
