Laboratory Techniques Flashcards
Quadrant Streaking Technique
Reduces or dilutes the number of cells using successive streaking zones on the petri plate and flaming the inoculation loop between zones
Motivation
The energizing of behavior in pursuit of a goal
Dilution Formula
(M1)(V1) = (M2)(V2)
Technological vs Biological Replicates
Technological: Same sample, different readings
Biological: Different samples
UAS - GAL 4 System
A binary expression system
GAL 4 comes from the yeast genome and is inserted into dorsophila and is regulated by an enhancer
In a separate fly, there is a UAS (Upstream activation sequence) that when GAL 4 is bound to it, expresses a reporter gene
The flies are crossed resulting in a fly with UAS - GAL 4 that has the ability to only produce GAL 4 in specific tissues allowing the reporter gene to be expressed
Binary Expression System
Any system that allows for the over expression or “knocking out” of a gene
RNAi
Used for gene regulation, RNAi will bind to a single strand of RNA forming double stranded RNA which will be destroyed
Hebbian learning
Neurons that fire together wire together so as the pre synaptic neuron fires and stimulates the post synaptic neuron the synapse improves.
En passant Synapse
In contrast to a normal neural synapse an En Passant synapse forms along the length of the axon and is usually only a temporary linkage
Start Codon
A codon that is recognzied by a ribosome to start translation of a genetic sequence
Stop Codon
Another specific codon that is recognized by the ribosome to stop translation.
