Lab Practical Flashcards
what are the 7 common bench top items?
1) Micropipette
2) Latex gloves
3) Face mask
4) Safety goggles
5) Bench top centrifuges
6) Vortex Mixer
7) Ethanol/ water spray bottle
What is the difference between Bench top centrifuge and vortex mixer?
1) Bench top centrifuge separates substances according to density
2) Vortex mixer mixes liquids in vials
What are the 4 waste disposal bins and what are the used for?
1) Sharps bin: disposal of sharp items
2) Biohazard waste bin: disposal of any waste that has pathogens that can be dangerous to people or environment
3) Normal waste bin: for general waste
4) Broken Glass bins: disposal of broken glass
What is the difference between Fume hood and laminar flow hood?
Fume hood protects you from any hazardous substances that could leak out while the laminar flow hood is to prevent the sample that your are working on from being contaminated
What are the 5 common symbols found in the lab?
1) Biohazard
2) Radioactive
3) Toxic (Skull sign)
4) Corrosive (Chemical with hand sign)
5) Flammable
what is the proper way to handle a microscope?
always use 2 hands one on the arm of the microscope and the other on the base of the microscope for support
how do you calculate the final magnification of a microscope?
you need to multiple the objective lens magnification by the eyepiece lens magnification (eyepiece lens always 10x)
What are the 5 care and maintenance steps for microscope?
1) never scratch the lens
2) always clean lens externally with lens tissue
3) always remove immersion oil immediately after use if not it will harden
4) remove any stain with moisten lens tissue
5) microscope should be clean and dry before put in box
What is the purpose of cell lysis solution in DNA extraction?
it is to break open the cell so than the organelles and everything inside can flow out
What is the purpose of Proteinase K in DNA extraction?
it is an enzyme that breaks down the protein structure
What is the purpose of RNase A in DNA extraction?
it is an enzyme that breaks down RNA material
What is the purpose of Protein precipitation solution in DNA extraction?
it is to pull all the protein debris together
What is the purpose of Isopropanol in DNA extraction?
it is to crystalize the DNA
What is the purpose of the cold buffer in chlorophyll extraction?
it is to prevent the cells from bursting
What is the purpose of the Acetone in chlorophyll extraction?
Because chlorophyll cannot dissolve in water thus we need to use acetone
