Lab 7 - Restriction Digest and Gel Electrophoresis of DNA Plasmid (pGLO) Flashcards
Agarose gel electrophoresis is a procedure used to separate DNA fragments. Based on what characteristic do the fragments separate?
molecular weight and shape
What are restriction enzymes?
A protein isolated from bacteria that cleaves DNA sequences at sequence-specific sites, producing DNA fragments with a known sequence at each end.
What are restriction enzymes used for in a laboratory context?
They are tools to analyse DNA and they create recombinant DNa for genetic engineering.
What are endonucleases?
Enzymes that break the phosphodiester bonds in nucleotides in the internal position.
What are exonucleases?
Enzymes that progressively digest the ends of DNA molecules.
What is the definition of ‘restriction modification’?
A protective system in which endonucleases are found in bacteria.
What are bacteriophages?
Viruses that infect bacteria.
What are recognition/restriction sites?
A specific nucleotide sequence recognized by enzymes.
What is special about typical restriction sites?
They are palindrome (read the same but backwards as foreward)
What are methylase and what are they used for?
An enzyme used to add methyl groups to the nucleotides of bacterial DNA.
