Lab 3: Staining Flashcards
acid fast bacteria are resistant to gram staining due to waxy cell surface caused by unusual complex lipids known as what?
mycolic acids
how do we overcome the problems of mycolic acids?
moderate heats and detergents to break down lipid components of call wall and allow dye penetration
carboxylic acid on mycolic acids react with _______ dye/
fushin
What are the three steps of acid fast stains?
- carbolfucshin stain allowed to penetrate into cell
- decolorizing agent applied
- counterstain added
What is used in acid fast staining to enhance penetration of carbolfucshion?
phenol
What is the counterstain added in acid fast staining?
methylene blue
What is the decolorizing agent used in acid fast staining?
acid-alcohol
What is the chemical makeup of acid-alcohol?
3% HCl and 95% ethanol
________: structured extracellular matriculates composed of polysaccharides?
capsules
Capsule producing strains often being more virulent. T or F?
T
Can capsule be stained though methods involving heat. T or F?
T
_______: stain the background but no the cells
negative stains
______: is precipitated onto the filament which increases diameter through coating the flagella
mordant
What do you call a cell that has a flagella coming out of one end?
monotrichous
What do you call a cell that has multiple flagella coming out of one end?
lophotrichous
What do you call a cell that has flagella coming out of both ends?
amphitrichous
What do you call a cell that has flagella extending from all sides?
petrichous
_______: dormant form of bacteria resistant to hostile environments
endospores
What is it about the endospore outer covering that causes the cells resistance to staining?
keratin
What is the effect of malachite green ?
forces the dye into the cell by heating
vegetive cells are decolorized with water and stained with __________________
spore safranin counterstain
________: the spore at the end of an endospore
terminal spore
_________: the spore at the almost end of the endospore
subterminal
__________: the spore is in the middle of the spore?
central
What are the steps of a flagella stain?
- get prepared slide
- place on staining rail
- break ampule dropper and apply it onto cell
- let sit
- Whirl pool bath
- Let dry for 45 min
- Look at flagella under microscope
What are the steps of acid fast stain?
- Create a mixed slide
- use toothpick to break up mixture
- heat fix
- add drops of carbolfucshin stain
- rinse
- continue rinsing and add 2-3 drops of acid-alcohol to decolorize
- counterstain with methylene blue
- rinse off excess with water
What are the possible results of the acid fast stain?
non acid fast bacteria will be blue/purple
acid fast bacteria will be red
what are the steps of an endospore stain?
- create a smear plate
- cover the slide with paper towel and add lactate green
- steam for 10 minutes and continue to add drops of malachite green
- remove paper towel and rinse
- counterstain with spore safranin
- rinse
- dry
what will the results of the endospore stain be?
there will be red vegetative cells and green endospores
