what is a hormone ?
is a signalling molecule that modulates activity of target tissue
what fluids could you use to measure reproductive hormones
milk - things are transferred from blood to milk
blood - serum or plasma
urine- metabolites
High progesterone =
cyclic cow in luteal phase or pregnant
low progesterone =
in follicular phase, not pregnant or non cyclic
What is LFIA?
is a paper based platform for the detection of hormones in body fluids, where the sample is placed on a test device and the results are displayed within minutes
LFIA Direct test
the target hormone is immobilized between two complementary antibodies like a sandwich-based assay. thus the presence of the test line indicates a POSITIVE result.
LFIA Competitive Test
the hormone blocks the binding sites on the antibodies on the test line, preventing their interactions with the coloured conjugate. therefore, a positive result is indicated by the lack of signal in the test line , control line should be visible regardless of result
What are PAGs?
pregnancy associated glycoprotein
they are secreted by the placenta (binucleate trophectoderm cells in ruminants)
What is an ELISA?
the ELISA is a plate-based assay technique designed for detecting and quantifying antigens
an antigen must be immobilized to a solid surface and then complexed with an antibody that is linked to an enzyme
