Where do we keep cultures and why?
In test tubes to prevent accidents and to avoid contamination of yourself and the environment.
What do we do when we spill cultures?
Cover with paper towels and saturate them with germicide for 15 minutes then dispose of it.
When handling contaminated material:
Don’t place contaminated instruments on bench tops, contaminated pipettes and cotton swabs must be disposed in biohazard container, never pipette by mouth, contaminated test tubes must be placed in wire basket in the autoclave bin, live mounts of hazardous organisms must be soaked in germicide for 15 minutes before washing with soap and water.
When using Bacti-cinerators:
Optimum sterilizing temperature (150F/15C) after 10 minutes. Don’t leave inoculating loops or needles in Bacti-cinerator unattended.
Labeling
Use wax pencils on test tubes and microscope slides.Permanent markers can be used on Petri plates.

Brightfield microscope
Contains two-lens systems for magnifying specimens: the ocular lens in the eye piece and the objective lens located in the nosepiece. Specimen is illuminated by light focused on it by substage (condenser).
What is the major limitation of a Brightfield microscope?
The absence of contrast between the specimen and the surrounding medium, which makes it difficult to observe living cells.
