Only antipsychotic shown to be more efficacious than others – However, it is only used for treatment resistant schizophrenia….

Side Effects of Particular Concern with Clozapine
Agranulocytosis : WBC and ANC monitoring required to dispense
Seizures at high doses
Constipation, Paralytic ileus
Sialorrhea
Myocarditis
Orthostatic hypotension and tachycardia
Monitor before each dose when starting, slowly titrate
1st antipsychotic
• In the presence of MAO inhibition, this drug or haloperidol increased catecholamine metabolites
• Mechanism of this effect via catecholamine (DA) receptor blockade
• Chemical, typical/first generation
• Low potency
