L19 + L20 Flashcards
Gompertz Function
When a tumor is composed of few cells, it may grow faster vs when a tumor is large, growth rate is slow due to less supply of O2 and nutrients
Tumor Growth Delay
Time after radiation that ir takes for the tumor to regrow to the same size at the start of XRT
CD133, CD 44, Nestin
In Vitro Assay
measures survival of cell lines under non physiological conditions
In Vivo Assay
Measures cell survival/tumor growth in animals
In Vitro clonogenic survival assay
Ability to form colonies
Survival is measured by plating cells and seeing how many colonies are formed
Plating efficiency
= % of colony formation with no dose
SF (Surviving fraction)
= % colonies (dose) / % colonies (no dose)
Spheroid Systems
must use a tumor cell line that grows in spheroids in cell culture
These cells are irradiated while intact in spheroid and separated into single cells for plating and colony formation.
tumor growth measurements
control animals have untreated tumors
experimental animals got RT
Primary end point
growth delay
(days for tumor to grow to X size after treatment) - (days for untreated tumor to grow to X size)
Tumor Lung Colony Assay
Uses tumors that form lung mets
inject tumor into mouse and wait for lung colonies to form and then count them
TCD50 tumor control assay
groups of animals with same tumor are treated with different doses of RT
Primary End point
Tumor control Dose 50 ( TCD 50)
Dose required to control 50% of tumors
Tumor limiting dilution assay
Transplantable leukemia in mice can be transmitted by intraperitoneal injection
