Investigation of stomatal numbers of leaves adapted to different enviroment COPY Flashcards
investigation of stomatal numbers of leaves adapted to different enviroments steps
make a replica of the epidermis
counting the stomata
calculate stomatal distribution
what are the steps to make a replica of the epidermis
place a fully expanded leaf on a white tile with its lower epidermis facing upwards.
stretch the leaf between two fingers of one hand with the other hand apply a colourless nail varnish between the veins
allow it to dry
apply a second layer of nail varnish
allow it to dry
hold a pair of fine forceps horizontally and insert one point between the epidermis and the nail polish layer
grip the layer and pull away maintaining tension
place replica on microscople
apply 2 drops of water
count the stomata
focus on the replica using x 10 objective and the refocus using the x40 objective. adjust the light intensity using the sub stage iris diaphragm to produce a suitable image.
count the number of stomata in the field of view. make a rule concerning stomata that are only partly in the field of view
count the number of stomata in nine more field of view
ensuring that no stoma is counted more than one
calculate a mean number of stomata in view
calculating stomatal distribution
from your microscope callibration you can calculate the area of the field of vie
calculate distribution of stomata where
mean number of stomata per mm =
mean number of stomata per field of view / area of field of view in mm2
