Describe how to observe cells in a prepared slide using light microscope
A
-Prepare slide.
-adjust coarse adjustment knob, bring stage closer to lens.
-Adjust fine adjustment knob until clear
-Adjust all lenses

Why do you use hydrochloric acid?
A
To soften and loosen the root tips, separate cells by breaking down the tissue which binds cell together

Why do you use mounted needle?
A
No air bubbles under the cover slip- no artifacts

Procedure
A
-Add 1M of HCL and heat in water bath
-Add root tip to HCL
-Remove and wash with distilled water
-Cut tip of root tip
-on slide
-stain sample

why press down firmly on cover slip?
A
-ensure cell is thin so that light can pass through

Why must you use a stain?
A
So that chromosomes are visible
