Enzyme action Flashcards
Aim
To investigate the effect of pH/temperature on enzyme activity, to identify optimum PH/temp and range in which enzyme works
Method
-Draw 3 Xs on 3 test tubes
-Add the milk solution to each test tube add water to one, indicates no enzyme action
-to 3 different add equal vol. of trypsin solution and ph7 buffer,
-place all 6 into water bath
-add a tube of trypsin and buffer to each tube and mix
-record for each to X to become visible
-calculate the mean
-repeat at different temps.
Rate of reaction= 1/mean
Safety considerations
-Broken glass and cutting
-Hot liquids and scalding
-Enzymes- allergies
-Hy
Methodology evaluations
-Subjective
-Water bath temp. may decrease
-Not enough intermediate Ph values to test for optimum
