Amphetamine - MOA
Inhibits norepinephrine storage in the sympathetic neuron. Increased cytoplasmic NE concentration causes reverse transport into the synapse by catecholamine transporter. Both in CNS and PNS
Amphetamine - effects
Vasoconstriction, cardiac stimulation, increased BP, CNS stimulation.
Tyramine - source and interaction.
May cause marked increased BP in pts MAO-inhibitors (e-g- for depression).
Found in bananas.
Cocaine - MOA
Inhibits catecholamine transporter in presynaptic neuron. Decreased NE reuptake, and increased synaptic NE concentration
Cocaine - effects
Vasoconstriction, cardiac stimulation, increased BP, blocks dopamine reuptake.
Cocaine - clinical uses
Local anesthesia
Cocaine - adverse effects
Ischemia and necrosis og nasal mucosa (abuse).
Severe hypertension.

Dopamine
Ephedrine
Psuedoephedrine
Ephedrine - MOA + effects
Activate α1- (vasoconstriction) and β-adrenoceptors (bronchodilation)
Ephedrine - adverse effects
Tachycardia. Hypertension. Urinary retention. CNS stimulation. Insomnia
Psuedoephedrine - special
Used illegally to make methamphetamine.
Psuedoephedrine - adverse effects
Tachycardia, urinary retention, CNS stimulation, insomnia.
