what is the structure of haemoglobin?
2π2π chains (foetal = π+π) + 4 haem molecules with iron atom
where is haem synthesised?
mitochondria - iron enters cell bound to transferrin -> vesicle -> mitochondria
where is globin synthesised?
cytosol
what is the structure of haem?
protoporphyrin ring with central iron atom (ferroprotoporphyrin), iron usually in ferrous Fe2+ form and combines reversibly with oxygen
which enzyme regulates rate of haem production?
d-ALAS (β aminolevulinic acid synthase)
how many functional globin genes are there?
8 arranged in 2 clusters - π cluster on short arm of Chr11 (π,,β&E genes) & a cluster (a&Cglobin genes) on short arm of Chr16
what are the normal adult haemoglobins?
HbA (π2π2) - 96-98%, HbA2 (π2d2) - 1.5-3.2%, HbF (π2π2) - 0.5-0.8%
