What is a genome?
Each genome is the sum total of an organism’s DNA and is expressed as the base sequence of the haploid set of chromosomes.
What is the purpose of the human genome project?
To identify and sequence the human genome. To provide a complete and accurate sequence of the 3 billion DNA base pairs that make up the human genome and to find all of the estimated 20,000 to 25,000 human genes.
What are the benefits of the HGP?
- diagnosis: data from the HGP provide more accurate diagnoses of inherited disorders
- prevention: identifying genetic factors that predispose some people to disabling conditions
- treatment
- human biology
- human evolution
What is a gene?
A section of DNA, with a unique sequence thats codes for a particular protein.
What is genomics?
Comparative study of the genomes of several species.
What is DNA made up of?
DNA is made up of molecules called nucleotides. Each nucleotide contains a phosphate group, a sugar group and a nitrogen base.
What is gene sequencing?
The identification of the exact sequence of nucleotides in a gene.
What is the function of a gene?
To code for the production of proteins.
What are the purines?
Adenine and Guanine. They have two rings.
What is a nucleotide?
The building block of DNA. Consists of a phosphate group, a base and a sugar.
What type of bonds are found between base pairs?
Hydrogen bonds.
What are the pyrimidines?
Cytosine and thymine. They have one ring.
What is the only direction that DNA can be replicated?
It can only be replicated in a 5’ to 3’ direction.
What are the requirements of DNA replication?
- original DNA template
- free dna nucleotides
- DNA polymerase
- Primers
What occurs during the first stage of DNA replication?
- dna is unwound by helicase
- dna primase binds to 3’
- dna polymerase builds off rna to create bases. Lagging strand replicates with multiple primers creating okasaki fragments
- exonuclease removes rna primase and replaces them with bases. Then proofreading occurs
- Ligase joins okazaki fragments
