Gel Electrophoresis Flashcards
Briefly outline the method for gel electrophoresis
1) Attach electrodes to ends of gel plate
2) Cut DNA sample into fragments
3) Place fragments in wells, closest to negative electrode
4) Apply current
5) Visualise bands using UV light and a fluorescent DNA dye
What are used to cut DNA sample into fragments?
Restriction endonucleases
Why does the DNA move towards the anode when a current is applied?
Because it is negatively charged due to the presence of phosphate groups
How do you visualise DNA bands?
UV light
Fluorescent DNA dye
What does the electrophoresis gel plate consist of?
Agarose covered in buffer solution
How does gel electrophoresis work?
Separates DNA fragments for identification/analysis based on their size
