What are the properties of basic functional groups (3)
A
- Groups that can accept protons
- Strong bases shift equilibrium to the right - large Kb small pKb
- Weak bases (don’t have much of a tendency to form bonds with protons) shift the equilibrium to the left - small Kb large pKb

What are the properties of amines (4)
A
- N acts as an H bond acceptor and if R = H, then it acts as an H bond donor
- high density of electrons on the nitrogen producing a polarity in the bond
- No R is ammonia; one R is a primary amine followed by secondary, tertiary and quaternary ammonium salt.
- Nitrogen is a bond acceptor which can react with water, producing an OH by-product.

What are the properties of aromatic amines (3)

- The aromatic ring is an electron-withdrawing group
- Aromatic amines have a pKa of 4.6, aromatic amines without the double bonds have a pKa of 10.6
- The lone pair is delocalised on the aromatic ring giving rise to resonance.

- When an acidic or a basic structure is present on a drug, only the non-ionised form crosses membranes
- Ka = [A-][H+]/[HA]
- pKa = log [HA]/[A-] + pH

Water solubility is pivitol for:
- drug formulation (e.g. preparing drug solutions)
- distrubution (e.g. the drug is transported by the blood)
- the formation of ammonium salts is advantageous too
