What is the practical for investigating the effect of PH on enzyme activity?

A

1 - put a drop of iodine solution into every well of a spotting tile.
2- Set up a water bath at 35 degrees.
3- Add some amylase solution and a buffer solution with a PH of 5 to a boiling tube.
4- Put the boiling tube in the water bath and wait for five minutes.
5- Add some starch solution to the boiling tube, mix and start a stop clock.
6- Every 30 seconds , take a sample from boiling my tube using a dropping pipette.
7- Put a drop of the sample into a well in the spotting tile.
8- Record how long it takes for iodine solution to stay browny orange. - all starch is used.

Repeat with buffer solutions at different PH values.
