Drugs in health and disease Flashcards
First generation antipsychotic drugs mechanism
Blocks dopaminergic receptors (D2)
- Non-selectively
Can also have cholingeric, alpha-adrenergic, histaminergic and serotonergic effects
First generation antipsychotic drug examples
Chlorpromazine
Levomepromazine
Promazine
Haloperidol
Effects from blocking dopaminergic receptors
Hyperprolactinaemia
Sexual dysfunction
Sedation (by decreasing arousal)
Side effects
- Extrapyramidal symptoms
- Parkinsonism: tremors, bradykinesia, muscle rigidity
- Tardive dyskinesea
- Increase in appetite due to loss of inhibition
- Acute dystonia
- Akathisia
Effects of blocking cholinergic receptors
Symptoms
- Dry mouth
- Constipation
- Micturition dysfunction
- Blurred vision
Effects of blocking alpha adrenergic receptors
Nasal congestion
Erection and ejaculation problems
Effects of blocking histamine receptors
Drowsiness
Effects of blocking serotonergic receptors
Hypotension
Temperature dysregulation
