Drugs for Dementia Flashcards
What is MOA of donepezil?
reversibly and noncompetitively inhibits centrally active acetylcholine estersase to increase Ach in the synaptic cleft for transmision
What is donepezil used for?
Alzheimer’s disease
Altered cardiac conduction (torsades, Brady cardia, heart block), N/V, diarrhea, rapid mood change with abrupt discontinuation, worsening PUD, asthma/COPD and bladder outflow obstruction are toxicities of what?
donepezil
What is MOA of memantine?
NMDA receptor antagonist
What is use of memantine?
Alzheimers disease
- confusion/dizziness/headaches
- severe skin hypersentivity
- altered mental status (hallucinations, delusions, insomnia, anxiety)
are toxicities of what?
memantine
What is rivastigmine?
reversible AChE inhibitor for dementia in transdermal patch
What is galantamine?
Reversible AChE inhibitor for dementia, nAChR
What is first line tx for AD?
AchE inhibitor, change type if not responsive or add memantine
What should be avoided for antidepressants in AD pts?
Tricyclics–> side effects and drug interactions
What is recommended for depression in AD pts?
SSRI–> citalopram or sertraline
What has limited efficacy and is associated with increased mortality?
Antipsychotic agents
