Droperidol Mechanism of Action
Acts centrally at sites where dopamine, NE, & serotonin act- alters CNS signal transmission, exerts antiemetic effect at red astrocytes in CRTZ great for motion sickness, moderate alpha adrenergic blocking ability. May occupy GABA receptors on postsynaptic membrane causing buildup of dopamine in intersynaptic cleft (cannot be utliized = contraindicated in Parkinsons patients), causes neuroleptic anesthesia
Droperidol CV Effects
Decreased: SVR, MAP Neutral: CO, Contractility Changes in QT (prolonged) may cause R on T phenomenon, torsades de Pointes & delayed repolarization
Droperidol Respiratory Effects
Minimal
Droperidol CNS Effects
Decreased CBF in dogs, no human data. Worsening of extrapyramidal side effects, contraindicated in Parkinsons
Droperidol Uses with dose
Antiemetic 0.625-1.25 mg IM or IV in adults, must have EKG 1 hour after giving.. give early!
