What is used in the dideoxy sequencing method?
A
Dideoxy NTPs (DDNTPs)
Same as DNTPs but no 3’ OH

Why doesn’t new synthesis of dna occur when ddtps are added to dna
A
Because there is a missing 3’ OH so new phosphodiester bonds can’t form

How does length vary when using ddtps
A
The length of dna fragment produced will vary depending on where the ddtp was incorporated
Shorter = close to start of sequence

How does gel electrophoresis show you the base sequence using ddtps
A
The fragments on bottom of gel are shortest meaning the sequence must be whatever ddtp base used eg cdtps so C must be first base
