The methodology steps
1) denature DNA with heat
2) make multiple copies of a segment
3) attach a primer
4) add polymerase, bases and terminator bases to a solution
5) grow complementary chains until terminator base
6) denature the growth chains
7) electrophorese the solution
how data is recorded
fragments run through a capillary gel electrophoresis
short fragments move quicker through pores of gel than long fragments
as each fragment crosses the “finish line” it is illuminated by laser so attached dye is detected
data recorded by detector as a series of peaks which correspond to DNA sequence
