DNA Replication - Lecture 1 Flashcards
How many base pairs located per turn of a DNA strand?
10
How long is each turn of DNA?
3.4nm long
What is conservative DNA replication?
A process of DNA replication that produces one fully original double helix and one fully synthesized double helix
What is dispersive DNA replication?
A process of DNA replication where two double helices are produced but each contain fragments of original and synthesized DNA
What is semiconservative DNA replication?
A process of DNA replication where two double helices are produced but each contained one strand of original DNA and one strand of fully synthesized DNA
How was it determined that DNA replicated semi conservatively?
- E.coli was cultured in a medium containing heavy nitrogen so that on all the DNA also contains heavy nitrogen
- This heavy DNA was then centrifuged have one heavy density band was observed
- Then light nitrogen was added to the medium and one hybrid of medium density was observed
- Later after several rounds of the centrifuge, mostly light nitrogen bands were observed with some hybrid bands
Who determined that DNA replicates semi-conservatively?
Meselon and Stahl
