Why is tissue often fixed is the tissue processing step.
Tissue is often fixed to preserve it in as life-like state as possible. Usually a chemical that cross links proteins such as formalin is used
What is staining
Staining is a process by which a tissue section is coloured.
How many main types of stains are there
- acidic stains
basic stains
neutral stains
What colour are acidic stains
The acidic stains are negatively charged and combine wit positively charged particles. They are red or pink in colour. example=eosin. tissue that stain red with acidic dye are termed acidophilic.
What colour are basic stains
Basic stains are positively charged, and combine with negatively charged particles. They are blue or dark purple in colour. example=Hematoxylin & toluidine blue are the commonly used ones.
