Different Types Of Microscopes Flashcards
How does a scanning electron microscope work?
•A beam of electrons is focused onto a specimens surface using electromagnetic lenses.
•Reflected electrons hit a collecting device and are amplified to produce an image on a photographic plate
What’s the sem magnification and resolution?
M= x 100,000
R= 3-10nm
How do transmission electron microscopes work?
Use a powerful beam of electrons to travel through a vacuum
What are drawbacks to using electron microscopes?
•Expensive
•not suitable to view whole cells and tissues
•black and white images
•non living only
•large
•training required
What’s the magnification and resolution of a tem?
M= x 500,000
R= 0.2 nm
What is the magnification and resolution of a light microscope?
M= x1500
R= 200nm
What do tem microscope produce?
An internal 2D image in black and white
What do sem microscopes produce?
External 3D black and white image of the cell
How does a confocal microscope work?
Uses a laser light to obtain image
Collects the image at different depths by scanning the laser through different focus positions
Allowing live specimens to be imaged without physical sectioning
