Steps you would take to obtain a pure culture from a mixed broth culture?

A
  1. wipe area with hypochlorite (javex) solution 2. light bunsen burner 3. pick up test tube of mixed cultures in dominant hand and inoculating loop in other hand 4. pass the loop through the blue part of the flame and let cool for 10 seconds 5. remove the cap of test tube with the same hand the loop is in 6. pass the top of test tube through flame 7. insert loop into tube, avoiding the sides, flame test tube again and recap 8. remove lid of agar plate and streak back and forth on 1/3 of plate 9. reflame loop and let cool 10. turn plate 90 degrees and streak again streaking into the previous streaks for the first couple 11. reflame loop and let cool 12. turn 90 degrees and streak again streaking into the previous streaks 13. replace lid of agar plate and invert it, label bottom of plate 14. place plate upside down on rack to be incubated
