Culture Media Flashcards
A semi-solid, non nutritive media with sodium thiosulphate to prevent oxidative changes to the organism
Maintains organism viability but does not support growth
Stuart’s Medium
Used for the collection of stool specimen for bacteriology studies
Buffered glycerol saline
used for the collection of stool for parastitology studies
5% Formalin
Chocolate tellurite is a selective medium, what is it used to culture ?
Corynebacterium species
This media contains the basic nutrients needed to provide growth of most organism
Nutrient Broth/Meat broth
Nutrient broth to which 2% agar has been added
Nutrient Agar
Heated sheep or horse blood to release extra nutients from the blood
Factor X and V
Chocolate Agar
This differentiates lactose fermenters from non-lactose fermenters and can be used to selective grow gram negative bacilli (enterobacteriacea) and inhibit gram positive cocci
MacConkey’s Agar
This selective medium is heated blood agar with CVNB (colistin, vancomycin, nystatin, bactrim) and isolates Neisseria Gonorrhea from genital swabs as it inhibits the growth of the normal flora
Thayer Martin agar
This medium is used to identify Salmonella, separate from Shigella species.
Salmonella will form yellow colonies which then turn pink with black centres
Shigella does not ferment the xylose so the colonies remain pink, and it also does not oxidize the thiosulphate to H2S so no black dots form
XLD
Xylose, lysine, deoxycholate Agar
Used for the isolation of bordetella pertussis
Bordet Gengou Agar
Used for the isolation of Corynebacterium diphtheria
Potassium tellurite is added, and is reduced by C.diptheria forming black colonies
tinsdale Agar/ Chocolate tellurite Agar
Used to demonstrate metachromatic granules of corynebacterium diptheria
Loeffler’s Serum Slope
This medium is used to isolate mycobacterium (M.tuberculosis)
It contains egg and an inhibitory green pigment dye to gram pos and gram neg bacteria
Lowenstein-Jenson Medium
This media suppresses coliform bacilli from faeces and enhances the growth of enteric pathogens
Selenite/Gram negative broth
