CRISPR/Cas9 system Flashcards
What is Cas9?
A nuclease with two active cutting sites that is able to cut both strands of dsDNA
What is tracrRNA?
The part of the sgRNA that is bound by Cas9
What is PAM?
Protospacer adjacent motif
- (Not sure what the function is, if you know feel free to add*)
- a DNA sequence immediately following the DNA sequence targeted by Cas9 in the CRISPR bacterial adaptive immune system. Cas9 will not successfully bind to the target DNA sequence if it is not followed by the PAM sequence (Wikipedia)*
What is sgRNA?
Synthetic guide RNA
It is a fusion of tracrRNA and spacerRNA
Where does the CRISPR/Cas9 system come from?
It is a prokaryotic immune system in the form of acquired immunity
What does Cas stand for?
CRISPR associated
How does the CRISPR/Cas9 system work?
Sorry guys, I can’t write it down now, but here’s a link to a really good video explaining both the discovery and the function of the system ;)
The presenter is the one who was part of the team developing the system.

What does CRISPR stand for?
cluster regularly interspaced short palindrome repeats
What is spacrRNA?
SpacrRNA is the part of sgRNA that binds the target sequence of DNA
