Depressants
drugs that reduce neuronal activity in the brain, target GABA receptor
GABA receptors
promote hyperpolarization of the postsynaptic cell
Orthosteric site
where GABA normally binds to the receptor, primary binding site
Allosteric modulators
change the functionality of the orthostatic site without competing for the same site
Barbituates
potent sedative-hypnotic drugs widely used in early 1900s, used to induce sleep in psychotic patients and to treat insomnia and anxiety
Long acting barbituates
low lipid solubility, slowly absored
Intermediate and short acting barbituates
moderate lipid solubility, absorbed faster with a 30 minute onset aciton
Ultrashort acting barbituates
highest lipid solubility, time to action mere minutes, lasting effects of about 30 minutes
REM suppression
reduces quality of sleep
REM rebound
reduces quality of sleep by repeated nightmares
GHB and GABA
each is a precursor and a by-product of each other
Sudden sniffing death syndrome
inhalants causing immediate arrhythmias and cardiac arrest
