Chromatography Flashcards
What is chromatography?
Method of separating mixture of soluble substance, e.g a mixture of dyes
What is the stationary phase and mobile phase?
Stationary phase - solid/liquid
Mobile phase - Liquid/gas
What happens to them both?
The components of the mixture dissolve in the mobile phase and move through the stationary phase
What is the distance they move determined by?
They distance they move is determined by how strongly they are adsorbed onto the solid stationary phase. i.e. components that adsorb strongly onto the solid stationary phase do not move far
What are the water and silica/alumina in the mixture?
Polar stationary phases
What are the two types of chromatography and what are the stationary and mobile phases?
Paper chromatography
Stationary - Water , Mobile - Solvent
Thin-layer chromatography
Stationary - Silica/alumina , Mobile - Solvent
What advantages does two-way chromatography have?
Allows more definite separation of a complex mixture
What are the stationary and mobile phase for gas liquid chromatography?
Stationary - Liquid or oil held in solid support
Mobile - Inert gas e.g. noble gas/nitrogen - gaseous sample mixed with this then passed through a coiled tube
