A
MOA- competitively inhibits dihydrofolate reductase. in higher doses may inhibit protein synthesis
-cannot supplement folate with this drug when using it to tx CA

tumor lysis syndrome
A
metabolic complications that occur during CA tx- esp with lymphoma & leukemia, Burkett’s, (ALL, AML)
- caused by the breakdown products of dying cancer cells
- can induce renal shut down
- complications- hyperkalemia, hyperphosphatemia, hypocalcemia, hyperuricemia, hyperuricosuria, uric acid nephropathy, acute renal failure
- prophylactic tx- oral or IV allopurinol(inhibits uric acid), adequate IV hydration to maintain a high urine output(>2.5L/day)
