cell fractionation Flashcards
1
Q
what is cell fractionation
A
process where cells are broken up and the organelles they contain are separated
2
Q
why is the tissue placed in a cold, buffered solution with the same water potential
A
cold - reduces enzyme activity that may break down organelles
buffered - so that pH does not fluctuate, which could alter the structure of organelles or affect the functioning of enzymes
same water potential - prevent organelles bursting or shrinking due to osmotic gain/loss of water
3
Q
what is homogenation
A
cells broken up by homogeniser, releases organelles from cell, resultant fluid (homogenate) is then filtered to remove any complete cells and large pieces of debris
