First step of washing c. elegans
use 1000 microliter M9 buffer 3x
run down plate to scoop c. elegans
Do we wash worms with pure water?
no
Second step of washing c. elegans
centrifuge test tube
make sure to balance the centrifugeq
How long should you centrifuge?
Really quick
a couple of seconds
Why not over-centrifuge?
Do not want to squish the worms
supernatant
liquid lying about the pellet after centrifuging
Third step of washing c. elegans
remove supernatant
add 1ml of M9 onto worms
flick the tube to resuspend the worms
Why do we wash?
Do not want to replate bacteria
want to wash bacteria and worm waste away
Fourth step of washing c. elegans
centrifuge again after flicking
and remove the supernatant
repeat
fifth step of washing c. elegans
resuspend in desired concentration
If worms are too concentrated?
Add more solution
If worms are not concentrated enough
re-centrifuge and remove supernatant
What concentration did we want?
Roughly 40 worms in 5 microliters

Hermatphoites
Eat bacteria (cheap)
40% same genes as humans
3 week lifesspan
scientists can manipulate the genome
