Biotechnology Flashcards
Where do enzymes cut?
Enzymes cut at a specific recognition sequence that contains a six-base palindrome.
What is a sticky end?
Sticky ends are the result when an enzyme cuts in different places on the recognition sequence
What is a blunt end?
A blunt end is the result when an enzyme is cut in the middle of the recognition sequence.
Explain the use of ligase
Sticky ends rejoin to complimentary sticky ends forming hydrogen bonds spontaneously, but to form the sugar-phosphate phosphodiester bonds you need an enzyme called ligase
What is a GMO?
a genetically modified organism is any organism that has had its DNA changed by geneticists in a lab.
What is a TGO?
A trangenic organism is a GMO that has been modified by adding genetic material from another species to its genome.
List the steps in recombinant DNA technology
isolating DNA from the donor and host organisms,
cutting the DNA using restriction enzymes,
joining the fragments with DNA ligase,
introducing the recombinant DNA into the host organism,
and selecting and screening transformed cells.
