Biologicals/Immunotherapies Flashcards
‘MAB’ refers to what kind of antibody?
Monoclonal antibody
‘LIM/IM’ refers to what kind of antibody?
Immune system
‘LIMO’ refers to what kind of antibody?
Murine (Mouse) Monoclonal antibody
‘LIXI’ refers to what kind of antibody?
Chimeric
‘LIZU’ refers to what kind of antibody?
Humanised
‘LIMU’ refers to what kind of antibody?
Fully Humanised
How does ‘Enlimomab’ work?
It is anti ICAM
- Block cellular adhesion molecules
- Blocks T cell migration, decreased IL-6
Chimeric Antibodies are…
Ones that have human sequences on Fc and F(ab) regions but murine ligand binding region.
Humanised Antibodies are…
Same as chimeric plus replace all the conserve regions within the ligand binding region
Only hypervariable regions are murine
Fully Humanised Anitbodies are…
Made my imunising transgenic mice carrying human Ig genes
or by in vitro recombination in bacteriophages
What are the main problems associated with soluble receptors?
- Rapidly cleared from circulation
- Homotrimeric TNF can still crosslink membrane receptors with soluble receptor bound
- May facilitate signal transduction by ligand passing
How can you fix the problem of soluble receptors causing signal transduction?
Use chimeric fusion proteins.
- Can bind 2 molecultes of TNFa which prevents interaction with receptor due to steric hinderance
What is the main problem associated with Etanercept?
Bone marrow supression
Which cytokines may be injected for anti-inflammatory cytokine therapy?
IL-4, IL-10 and IL-13
When are immunotherapies usually prescribed?
Only after failure with DMARDs
