To break down cell membranes, releasing contents of cell.

To break down proteins associated with DNA (e.g. histones)

- Stopper flask (e.g. with cotton wool)
- Use sterile broth
- Disinfect surfaces
- Have bunsen flame nearby to create upward air flow.
