what is aseptic technique?
is a process used to achieve asepsis to prevent the transfer of potentially pathogenic micro-organisms to a site that may cause infection
why do we need aseptic technique?
prevents spread of pathogens
when is aseptic technique used?
- handling surgical equipment
- sterilisation of work surfaces
- reducing contamination from the environment
- prevent studied organisms being contaminated
- helping baby birth by vaginal deliver
how do you maintain aseptic conditions in a lab?
- use of airflow cabinets
- using sterile loops when transferring cultures
- sterilising or disposing used equipment
- washing hands thoroughly
what does sterile mean?
free from micro-organisms

- cell and tissue culture
- prep of medical test kit

how do you streak a plate using aseptic technique?
- sterilise the loop in the Bunsen burner until it turns red
- let the loop cool and take an isolated colony from the agar plate
- spread the isolated colony on the first quadrant of the petri dish
- put the loop and pack away
- flame the loop and pack away
- incubate the plate for 24 hrs
what is contaminated culture in biology?
a culture in which bacteria from a foreign source have infiltrated the growth medium
what is plant tissue culture?
is a collection of techniques used to maintain or grow plant cells, tissues or organs under sterile conditions
why is contamination bad?
- cause experiments to be repeated, resulting in delays
- costly wastage
what is the cauliflower cloning method?
- place forceps in sterilise solution called SDICN
- clean the bench and wipe area
- collect a cauliflower and place it on a petri, using a scalpel and cut it into a smaller part
- put the cauliflower in a jar. every 2-3 mins, swirl the jar gently. repeat for 15 mins
- strain the liquids and put the cauliflower into a agar growth medium
- label the petri dish and incubate in a warm lab
what are potential errors that can occur while streaking a plate?
- large amounts of inoculum used
- heavy streaking, agar is gouged
- drops of moisture on the plates surface
- loop not flames
what is a growth medium?
is a solid, liquid or semi-solid designed to support the growth of a population of micro-organisms or cells. is often specific to the bacteria
what are agar plates?
petri dish that contains a growth medium solidified with agar to culture micro-organisms
